Anti-Stress, Behavioural and Magnetoencephalography Effects of an L-Theanine-Based Nutrient Drink: A Randomised, Double-Blind, Placebo-Controlled, Crossover Trial

This rigorous double-blind, placebo-controlled crossover study in 34 healthy adults (18–40 years) tested an L-theanine-based drink (200 mg L-theanine) against placebo using a demanding cognitive multitasking stressor. Subjective stress response to the stressor was significantly reduced one hour post-dose in the active group (p=0.003), and salivary cortisol — the primary stress hormone from the HPA axis — was significantly reduced three hours post-dose (p=0.047). Brain imaging via magnetoencephalography (MEG) showed elevated posterior alpha oscillatory activity in those with higher trait anxiety after treatment, consistent with L-theanine's proposed mechanism of promoting a relaxed-but-alert brain state. Critically, L-theanine achieves anxiolytic effects through multiple neurochemical routes — increasing brain GABA and dopamine, inhibiting glutamate reuptake, and stimulating alpha-wave activity — all without the sedation associated with pharmaceutical anxiolytics.

Dietary Supplementation with Lactium and L-Theanine Alleviates Sleep Disturbance in Adults: A Double-Blind, Randomised, Placebo-Controlled Clinical Study

This 8-week RCT in 40 adults with sleep disturbance (PSQI > 5) tested a combined Lactium and L-theanine supplement against placebo. Within the L-theanine group, significant improvements were observed in total sleep time, sleep efficiency, sleep onset latency, and reduced wake-after-sleep-onset counts over the 8 weeks. Gut microbiome analysis also revealed significant increases in Ruminococcus and Blautia — bacteria associated with better sleep quality — in the supplemented group. The authors attribute L-theanine's sleep benefit to modulation of inhibitory neurotransmitters (GABA and glycine) and excitatory neuron suppression, promoting restful sleep without inducing grogginess. No adverse events were reported, and liver function markers remained normal throughout.

The Association Between Vitamin B6 and Anxiety

This review and the subsequent landmark RCT published in 2022 (Tarleton & Waddell, Human Psychopharmacology) establish the mechanistic and clinical rationale for vitamin B6 in anxiety and mood. B6 (pyridoxine) is an essential cofactor in the biosynthesis of GABA, serotonin, and dopamine — meaning it is a rate-limiting upstream factor for the very neurotransmitters that regulate calm and mood. Deficiency in B6 is associated with reduced GABAergic activity and increased neuronal excitability, which manifests as heightened anxiety and irritability. A 2022 double-blind RCT (Tarleton et al.) in young adults found that 100 mg/day of vitamin B6 for one month significantly reduced anxiety scores and suppression of visual evoked potentials — a marker of cortical excitation — consistent with enhanced GABAergic inhibition. The 6 mg dose in this supplement is within the standard dietary reference range and supports normal neurotransmitter synthesis, though substantially below the doses showing acute anxiolytic effects in human trials.
