This systematic review analyzed 18 published case reports and clinical trials on biotin supplementation for hair and nail changes. Every single patient who received biotin had an underlying pathology — such as brittle nail syndrome, uncombable hair syndrome, or inherited biotin metabolism defects — and in all 18 cases, clinical improvement was documented after supplementation. Importantly, the authors conclude that while biotin supplementation is clearly effective when a deficiency or relevant pathology exists, there is currently insufficient evidence to support its use in otherwise healthy individuals for cosmetic hair or nail enhancement.

This comprehensive review, covering 125 published studies, examines every major micronutrient's role in the three most common forms of non-scarring hair loss: androgenetic alopecia, telogen effluvium, and alopecia areata. Serum Vitamin D Levels and Alopecia Areata: A Case-Control Study

This hospital-based case-control study enrolled 135 clinically diagnosed alopecia areata patients and 135 age- and sex-matched healthy controls, measuring serum 25(OH)-vitamin D levels in all subjects. Patients had significantly lower mean vitamin D levels than controls (22.87 vs. 27.34 ng/mL, p=0.0004), and — critically — vitamin D levels showed a strong negative correlation with disease severity: the more extensive the hair loss, the lower the vitamin D level. Patients with the most severe forms (alopecia totalis and universalis) had the lowest vitamin D levels of all, and patients with nail changes associated with alopecia areata also had significantly lower vitamin D than those without nail involvement.

Vitamin D Status and Efficacy of Vitamin D Supplementation in Atopic Dermatitis: A Systematic Review and Meta-Analysis

This rigorous meta-analysis pooled data from 7 observational studies (986 AD patients, 657 controls) and 4 randomized double-blind, placebo-controlled trials to assess vitamin D's role in eczema (atopic dermatitis). Patients with atopic dermatitis had significantly lower serum vitamin D levels than healthy controls across all ages, with the deficit most pronounced in children. Crucially, vitamin D supplementation across the 4 clinical trials significantly reduced eczema severity scores (both SCORAD and EASI measures), with the proposed mechanism being that vitamin D boosts the antimicrobial peptide LL-37 in the skin, strengthening the barrier against pathogens — a key vulnerability in eczema sufferers.
